SS-31 (Szeto-Schiller peptide 31), also known as Elamipretide, is a mitochondria-targeted tetrapeptide with the sequence D-Arg-2'6'-dimethylTyr-Lys-Phe-NH2. It is selectively concentrated in the inner mitochondrial membrane where it interacts with cardiolipin, a critical phospholipid essential for mitochondrial electron transport chain function. Mechanism of Action
SS-31 localizes to the inner mitochondrial membrane via electrostatic and hydrophobic interactions with cardiolipin. By stabilizing cardiolipin, it preserves the structure of cristae and supports optimal electron transport chain (ETC) complex assembly. SS-31 scavenges reactive oxygen species (ROS) at the mitochondrial membrane interface, inhibiting cytochrome c peroxidase activity and reducing oxidative damage. This mechanism supports mitochondrial membrane potential (ΔΨm) and ATP synthesis in cellular research models.
